Abstract

[en] Racemic ketamine receives breakthrough therapy designation for major depressive disorder with imminent risk for suicide, from the US Food and Drug Administration. (R,S)-Ketamine was synthesized in 1962 by the American chemist, Calvin Lee Stevens. Until to date, this pharmaceutical product was used as anesthétique in the medicinal field. Racemic ketamine is synthesized according 4 general steps under batch conditions. However, the poor efficiency and flexibility of batch processes is one out of several factors leading to drug shortages reported by the FDA. In this research, we showed the investigation of racemic ketamine synthesis in order to develop innovative strategies for the preparation under continuous-flow process and purification of racemic ketamine by crystallization process.
