Abstract

[en] This master thesis aims to describe the development of an analytical workflow of an in vitro model for gastrointestinal epithelial cells oxidized using a chemical agent, hydrogen peroxide. Three cellular lines, namely Caco2, HCT116 and HT29 were cultured and oxidized using H2O2. Media resulting from oxidation was collected and analyzed using multidimensional gas chromatography coupled to time-of-flight mass spectrometry (GCxGC-TOFMS). Data collected was analyzed for each cell line using multivariate statistics such as PCA to differentiate controls from oxidized cells. Several VOCs were identified as potential biomarkers already highlighted by other studies, namely hexanal and glutaric acid, butyl isobutyl ester.
